Curcumin Reduces an Adhesion Molecule Expression by Inhibiting STAT3 Phosphorylation in Vascular Endothelial Cell

Abstract
Purpose: We explored whether curcumin, a flavonol found in many fruits and vegetables, can modulate expression of intercellular adhesion molecule (ICAM-1) and vascular cell adhesion molecule 1 (VCAM-1) proteins in human umbilical vein endothelial cells (HUVECs) treated with lipopolysaccharide (LPS), and investigated the mechanism underlying this modulation.

Materials and Methods: Levels of each adhesion molecule, nuclear factor kappa B (NF-kB), and signal transducer and activator of transcription 3 (STAT3) were determined using Western blot analysis. The effect of curcumin on NF-kB promoter activity was determined using an NF-kB luciferase reporter assay. The effect of curcumin on monocyte binding to endothelial cells was determined using a THP-1 cell (human acute monocytic leukemia cell line) monocyte adhesion assay.

Results: Curcumin blocked LPS-mediated ICAM-1 and VCAM-1 up-regulation in HUVEC cells. Curcumin did not affect LPS stimulated IkappaB¥á (IkB¥á) degradation, but did reduce LPS-induced STAT3 phosphorylation. In a luciferase reporter assay, curcumin decreased NF-kB activity. Curcumin also reduced adhesion to HUVEC cells by LPS-stimulated human acute monocytic leukemia (THP-1) cells.

Conclusion: Curcumin inhibits ICAM?1 and VCAM?1 expression on vasculature walls by reducing STAT3 phosphorylation and NF?kB binding to DNA.
